TIM-3 belongs to the T-cell immunoglobulin and mucin domain (TIM) family and is often referred to as CD366. Expressed by T-cells, it suppresses the effector function of cytotoxic T lymphocytes and Th1 cells and triggers cell death upon interaction with its ligand, galectin-9. TIM-3 is an important immune checkpoint in cancer, often expressed at a high level on exhausted T-cells in tumours, meaning that multiple clinical trials are ongoing to evaluate the efficacy of various TIM-3 inhibitors. TIM-3 is also studied by researchers investigating autoimmune diseases such as multiple sclerosis, type I diabetes and SLE.
