Recombinant Antibodies

An overall summary of recombinant antibodies and their advantages when used in scientific research.

Recombinant antibodies can be produced as either mono or multiclonal and are produced using in vitro cloning. This means that there are no animals required within the manufacturing process indicating that not only is less time required in order to synthesize and obtain the antibody, it also avoids any ethical issues than with traditional antibodies. Traditional methods utilise animals’ biological systems which results in a lack of control over the sequence of proteins produced. The synthesis of recombinant antibodies demonstrated in figure 1 and begins at the source cells where the antibody genes are recovered, once complete, the genetic material is amplified and cloned into a gene appropriate for a phage vector. Once the genetic material is modified in the vector, it means that a genetically altered version, commonly of E .coli, is able to infect the body, spreading altered genetics to host cells where an exchange of genetic material takes place resulting in the host cell producing the desired antibody.

There are a variety of ways that recombinant antibodies can be used in research, medicine, and life sciences. These include therapeutics against cancer, autoimmune diseases, and infections, diagnostic tools in lateral flow test kits, ELISA, and a range of immunoassays. Additionally, recombinant antibodies play a profound role in molecular biology for example western blots, immunofluorescence, flow cytometry, and immunohistochemistry all of which can be done faster and more reliably as a consequence of the antibodies being recombinant.

Monoclonal antibodies have a plethora of advantages and applications. Recombinant monoclonal antibodies consist of identical antibody chains that help increase reproducibility as well as provide a guaranteed supply, improved sensitivity, and confirmed specificity. These attributes make monoclonal antibodies best suited for the synthesis of recombinant antibodies as opposed to aiming to replicate the variable effect of polyclonal antibodies.

There is a range of advantages that the utilisation of using recombinant antibodies can provide, the first of which is the high level of consistency between batches, this is undertaken by using a controlled production line with a unique set of genes and results in greater reliability and validity within its range of applications. Another advantage is recombinant antibodies' improved sensitivity and high affinity demonstrated by the varying batches' Kd value. The Kd value is the measurement of the affinity where the lower the number, the higher the affinity of the antibody, recombinant antibodies produce a result of 4.7E-11 at the higher end compared to standard antibodies higher end being approximately 10-9. Another significant advantage of recombinant antibodies is the opportunity to engineer a specific application into the final product resulting in individuals being able to get the exact desired antibody for their research or application. Additionally, is the ease of scale since they rely on antibody genes in expression vectors and can therefore be carried out at any scale, this, plus the aforementioned factors demonstrate that they are a great solution for long-term studies.
