dsRNA antibodies

dsRNA Antibodies were first generated to indicate viral infection of plants, but as viral vectors are being increasingly investigated for gene and cell therapy, the use of these antibodies offer a useful tool to enable high throughput testing of residual nucleic acids by ELISA, 1 but there are a number of clones available on the market, so which is best for your experiment? 

Some clones are available as recombinant antibodies, and may be offered in a range of isotypes and formats. Do contact us if you have specific requirements in mind.

J2 ( Mouse IgG2a, Kappa + recombinant) Species independent

This recombinant mouse monoclonal antibody, rJ2, is useful for detecting double stranded RNA of greater than 40 bp in length which is generated during the replication of positive sense genome viruses. It works with HCV, rhinovirus, dengue virus, and others. It does not work for dsRNA produced by influenza A virus and other negative sense genome viruses. Often regarded as the gold standard clone. Recently J2 has also been used to monitor the removal of dsRNA from in vitro synthesised mRNA preparations that may have potential use in gene therapy (Kariko et al., 2011).

9D5 Mouse IgG1 recombinant Viral

 

Detection level of dsRNA by using the 9D5 antibody remained low in comparison to K1 and J2 antibodies (Störk et al., 2021; PMID: 34921222). However, 9D5 was found to be more sensitive than the J2 and K1 for the detection of dsRNA from negative-stranded and ambisense RNA viruses, as well as single-stranded DNA viruses in IF analysis (Son et al., 2015; PMID: 26136565 and Mateer et al., 2018; PMID: 30087859)

P6 (Mouse IgG3+ Recombinant) Species independent

 

This antibody is recommended for the detection and analysis of ssRNA and ds RNAs. It can be also used to research new anti-viral strategies. The Light chain of P6 has similar heavy chain to K1 & J2, however the light chain sequence is unrelated.

Suggested applications: functional assay, ELISA

1D3 (Rabbit + Recombinant) - Virus 

 

 

The antibody binds specifically to ds-RNA, which is part of the genetic material of some viruses, such as rotaviruses and the bluetongue virus. The antibody can be used as a diagnostic tool, to determine the nature of an unknown pathogen, and could potentially be used in gene therapy.

K2 (Mouse IgM Kappa)

 

Anti-dsRNA monoclonal antibody K2 recognises double-stranded RNA (dsRNA) provided that the length of the helix is greater than or equal to 40 bp dsRNA. Recognition is independent of the sequence and nucleotide composition of the antigen. All naturally occurring dsRNAs investigated up to now (40-50 species) as well as poly(I)-poly(C) and poly(A)-poly(U) have been recognised by Anti-dsRNA monoclonal antibody K2.

As described by Schönborn et al. (1991), Anti-dsRNA monoclonal antibody K2 binds with high avidity to all dsRNAs investigated. Isotype alternative to K1 & J2, however as it may aggregate at high concentrations it can not be lyophilized and may require shipping on ice.

J5 Mouse IgG2b kappa

 

J5 IgG2b antibody recognizes dsRNA with very similar affinity and specificity to J2 but has a different isotype , however with recombinant versions of J2 also available the market of J5 has diminished.
